An excellent local example of Gothic Revival architecture, St. Mary's Church also has a long history as Uxbridge's oldest Catholic parish. Begun in 1850, when Reverend Patrick McGrath of Hopkinton came occasionally to Uxbridge to conduct religious services in the houses of farm workers on the estate of Joseph Thayer, St. Mary's was made a parish in August of 1853. In 1852 or 1853, the Reverend Charles O'Reilley purchased a lot at 66 North Main Street, where construction of the original St. Mary's Church was completed in 1855. At the time of it's completion, the origina l St. Mary's Church served a parish which included the towns of Grafton, Millbury, Northbridge, Douglas and Uxbridge.

Around 1925, the parish purchased the Mendon Street site of the present church. The construction of the new church was completed by August 15, 1926 when the building was dedicated. At approximately the same time, the original St. Mary's Church was sold, heavily altered by it's new owners and re-opened as the Bijou Theatre on October 22, 1926. Since 1926, the Mendon Street church of St. Mary's has remained in continuous use by it's original congregation.
