La ermita fue construida a las afueras de la ciudad bajo la advocación del Apóstol Santiago, en torno al siglo XV. Fue su patrón Diego de Guzmán y Quesada, caballero de la Orden de Santiago, quien ordenó la construcción de una cripta para sus caballeros. De su testamento se deduce que la ermita fue reedificada en 1631. Nuevas obras de mejora se realizan en la década de 1730.

El edificio se libró de la destrucción que provocó en la ciudad el Terremoto de Lisboa. Sin embargo, entre 1787 y 1794 debe ser reconstruido de nuevo, bajo probables trazas de Antonio de Figueroa. En 1843 se instaló un órgano que no ha llegado hasta nuestros días.

Desde su construcción ha tenido diferentes usos además del litúrgico. Así se convirtió en el primer centro docente de enseñanzas medias y superior de la ciudad al ser sede en el siglo XVII de las cátedras de Latinidad y Gramática creadas por Diego de Guzmán y Quesada.? Con motivo de la epidemia de cólera morbo de 1854, la ermita es cerrada al culto para ser transformada en hospital, pasando su ajuar litúrgico y artístico a la cercana Parroquia de San Pedro. En 1869 pasa a manos del Estado y en 1883 al Ayuntamiento de Huelva convirtiéndose en asilo y escuela.? Para esta función fue necesario dividir el espacio en dos pisos. A mediados del siglo XX fue local de ensayos de la banda municipal de música, hasta su cierre por ruina.

Es reseñable decir también que se convirtió en el lugar donde se refugió al poeta Miguel Hernández en su huida hacia Portugal tras la Guerra Civil española.

La reapertura del edificio, recuperando su uso religioso, tiene lugar en 1995, tras las obras de restauración dirigidas por Alfonso Martínez Chacón. Tras estas obras vuelve a residir en el templo la Hermandad del Santo Entierro, cuya titular mariana proporciona a la ermita su advocación.

La ermita es de una sola nave con capilla mayor acabada en testero plano bajo la que se encuentra la cripta de los caballeros de Santiago, del siglo XVII. Su fachada principal, en el lado de la Epístola, cuenta con una portada rematada por espadaña. Responde a la tipología barroca popular de finales del siglo XVIII. En 2015 se hicieron obras de ampliación de la puerta de la nave del Evangelio, con objeto de facilitar la salida de los pasos procesionales.

The hermitage was built on the outskirts of the city dedicated to the Apostle Santiago, around the 15th century. It was his patron Diego de Guzmán y Quesada, a knight of the Order of Santiago, who ordered the construction of a crypt for his knights. From his will it can be deduced that the hermitage was rebuilt in 1631. New improvement works were carried out in the 1730s.

The building was spared from the destruction caused in the city by the Lisbon Earthquake. However, between 1787 and 1794 it must be rebuilt again, under probable traces by Antonio de Figueroa. In 1843 an organ was installed that has not survived to this day.

Since its construction it has had different uses besides the liturgical one. Thus, it became the first teaching center for secondary and higher education in the city, being the headquarters in the 17th century of the chairs of Latinity and Grammar created by Diego de Guzmán y Quesada. On the occasion of the morbid cholera epidemic of 1854, the hermitage is closed to worship to be transformed into a hospital, passing its liturgical and artistic trousseau to the nearby Parish of San Pedro. In 1869 it passed into the hands of the State and in 1883 to the Huelva City Council, becoming an asylum and a school. For this function it was necessary to divide the space into two floors. In the middle of the 20th century it was a rehearsal place for the municipal music band, until its closure due to ruin.

It is also noteworthy to say that it became the place where the poet Miguel Hernández took refuge in his flight to Portugal after the Spanish Civil War.

The reopening of the building, recovering its religious use, takes place in 1995, after the restoration works directed by Alfonso Martínez Chacón. After these works, the Brotherhood of the Holy Burial returns to reside in the temple, whose Marian owner provides the hermitage with its dedication.

The hermitage has a single nave with a main chapel finished in a flat front, under which is the crypt of the Knights of Santiago, from the 17th century. Its main façade, on the Epistle side, has a doorway topped by a belfry. It responds to the popular baroque typology of the end of the 18th century. In 2015, works were carried out to enlarge the door of the Gospel nave, in order to facilitate the exit of the processional steps.
