O presente edifício, um dos maiores da diocese, foi erguido no estilo barroco. João Gomes Coelho, mestre pedreiro de Lisboa, dirigiu as obras de construção com relativa rapidez. No ano de 1710, é a vez do mestre pedreiro Domingos Gonçalves assegurar a finalização da cobertura em abóbada.

Como afirma Túlio Espanca, a volumetria do templo e a sua monumentalidade interior, sumptuária, pode admitir também um patrocínio dado aos religiosos vicentinos pelos Gamas (o condado não só albergava a Vidigueira, como ainda Vila de Frades). O seu altar-mor é mesmo considerado um dos mais belos na região do Alentejo, atestando assim o inegável esteticismo desta Igreja, que possui, igualmente, quatro altares laterais.

Este templo apresenta algumas esculturas antigas: São Cucufate (padroeiro da Paróquia retratado por uma imagem seiscentista que antes estava no Mosteiro da antiga villa romana), Nossa Senhora da Assunção (lavrada por volta de 1700), Nossa Senhora da Conceição (obra dos finais do século XVII), São Pedro (escultura do século XVII), São Sebastião (imagem seiscentista e padroeiro da desaparecida ermida titular desta designação), o Menino Jesus, etc.

Recorde-se ainda que se encontraram algumas sepulturas no corpo da nave desta Matriz. Supõe-se que uma delas seja a do poeta João Xavier de Matos, falecido, em Vila de Frades, a 3 de Novembro de 1789. A maior parte dos epitáfios aí presentes parece ser praticamente indecifrável.

Esta majestosa Igreja Matriz encontra-se no centro da Vila, mais concretamente no Largo de São Cucufate. Não se encontra regularmente aberta ao público, mas pode ser visitada por prévia marcação junto do Pároco local, ou na Junta de Freguesia tel: 284441762.

Its altar is even considered one of the finest in the Alentejo region, thus confirming the undeniable aestheticism of this Church, which has also four side altars.

This temple has some ancient sculptures: St. Cucufate (parish's patron saint portrayed by a seventeenth-century image that was previously in the Monastery of the ancient Roman villa), Our Lady of the Assumption (drawn around 1700), Our Lady of Conception (work of the late XVII century), St. Peter (sculpture of the seventeenth century), San Sebastian (seventeenth-century image and patron of the missing hermitage holder of this title), the Child Jesus, etc.

It is further recalled that some graves found in the body of the ship of this Matrix. It is assumed that one of them is the poet John Xavier de Matos, who died in Vila de Frades, 3 November 1789. Most of the epitaphs present there appears to be virtually unreadable.
